Can I use Microsoft's Data Publishing Wizard to script all objects in my SQL 2005 database to a .SQL file & then use this file to create all the object & data in my new SQL 2008 database using myLittleTools?

Yes this is something you can do.
Even if the best solution would be that your hosting company provides myLittleBackup ( http://www.mylittlebackup.com )

How do I execute the .sql file? I tried cutting & pasting the file con tents to myLittleTools query window but the file is 100mb & it didn't work?

100MB?!!
Do not waste your time it won't work...
The only solution is to restore a backup file. Contact your webhoster for that.
